Omega Biotek's RNA Solution Based kit, E.Z.N.A.® SQ Total RNA Kit, is designed for isolating total RNA from animal and plant tissue and cultured cells in under 60 minutes. It's protocol removes contaminants and enzyme inhibitors, such as proteins and divalent cations. RNA purified using the E.Z.N.A.® SQ Total RNA method can be directly used for applications including RT-PCR, Northern Blotting, NPA's (nuclease protection assays) and other enzymatic reactions.
This solution based system can be easily scaled up and down based on the starting material. There is no need for phenol/chloroform extractions, and time-consuming steps such as CsCl gradient ultracentrifugation that are eliminated.
The principle of the SQ Total RNA kit uses a highly efficient solution based system to provide a convenient, fast, reliable and non-toxic method to isolate high quality RNA from various samples. Samples are first lysed with RCL Buffer. Cellular proteins and genomic DNA are removed by precipitation with PNP Buffer while RNA will remain in solution. RNA is further purified by isopropanol precipitation.
Yield of total RNA purified using the E.Z.N.A.® SQ Total RNA Kit. Purified total RNA from 50 mg rice leaf was isolated with the E.Z.N.A.® SQ Total RNA Kit. RNA was eluted in 30 µL DEPC-treated water and quantitated using Thermo Fisher’s NanoDrop® 2000.
Total RNA purified using the E.Z.N.A.® SQ Total RNA Kit. Purified total RNA from 50 mg plant leaf samples was isolated with the E.Z.N.A.® SQ Total RNA Kit. RNA was extracted from plant samples as follows: sample 1 from rice leaf, sample 2 from corn leaf, sample 3 from soybean leaf, sample 4 from tomato leaf, sample 5 from corn seeds, and sample 6 from soybean seeds. Total RNA (5% of total purified RNA) was analyzed on a 1% agarose gel to demonstrate yield and quality of the RNA.
