DNA Recovery from Polyacrylamide Gels
The E.Z.N.A.® Poly-Gel DNA Extraction Kit provides a fast and efficient method to purify the single- or double-stranded DNA fragments from polyacrylamides gel. Following the PAGE, the DNA fragments are first visualized by UV shadowing or ethidium bromide staining. The gel slice contains DNA bands of interest that are cut and incubated with a special elution buffer to elute DNA from gel. Polyacrylamide gel is efficiently removed with poly-gel filter column. The sample contains are mixed with a special formulated buffer and loaded into the HiBind DNA spin column. After two quick washing steps, purified DNA fragments can be eluted from HiBind DNA spin column with low salt Elution Buffer or water. Purified DNA is ready for downstream applications such as sequencing, ligations, PCR amplification, restriction enzyme digestion.

The Poly-Gel DNA Extraction (Polyacrylamide) combines the simplicity of PAGE with the technology of HighBind silica matrix spin columns to allow rapid and consistent recovery of ssDNA and dsDNA from acrylamide gels. DNA bands, from 50bp to 40kb, have recoveries up to 85% and free of contaminants. Following PAGE, oligonucleotides are first visualized by UV shadowing or by ethidium bromide staining and then eluted for 1-4 hours using EDTA-containing buffer to eliminate degradation of DNA by contaminating DNase. A specially formulated buffer is then added and the whole mixture is passed through a HighBind DNA EZ-SpinKit column. The bound DNA is finally washed to remove salts, buffers, and any protein contaminants, and then eluted with sterile deionize water or low salt buffer. Organic extractions and alcohol precipitations are eliminated reducing sample handling time and increasing yields. Furthermore, the spin column format allows parallel processing of multiple samples. The purified DNA is suitable for PCR, ligation reactions, hybridization techniques, and other procedures.
